Standardin ISO 13053 tässä osassa kuvataan Six Sigmana tunnettu liiketoiminnan kehittämismenetelmä. Menetelmä koostuu yleensä viidestä jaksosta: määrittely, mittaus, analysointi, parantaminen ja ohjaus (joiden englanninkielisistä vastineista muodostuu lyhenne DMAIC). Standardin ISO 13053 tässä osassa suositellaan parhaita käytäntöjä kuhunkin Six Sigma -projektin toteuttamisen aikana käytettävään DMAIC-menetelmän jaksoon. Lisäksi neuvotaan, kuinka Six Sigma -projekteja olisi johdettava, ja kuvataan näihin projekteihin osallistuvan henkilöstön roolit, asiantuntemus ja koulutus. Standardia voidaan soveltaa sekä valmistusprosesseja että palvelu- ja kaupankäyntiprosesseja käyttäviin organisaatioihin.
